How to Store Peptides: Best Practices
Temperature Control
Temperature is the single most important variable in peptide storage. Most peptides remain stable for extended periods when stored at -20°C or below. For long-term storage, -80°C is ideal for most compounds.
Short-term storage (up to 2-4 weeks) at 4°C can work for many peptides, but extended refrigeration increases degradation risk. If you're working with peptides that will be stored for more than a month, freezer storage is essential.
When storing peptides at low temperatures, allow the vial to come to room temperature before opening. Opening a cold vial when warm air contacts the cold surface can cause condensation inside the vial, introducing moisture that degrades peptides. A simple 15-20 minute equilibration at room temperature before opening prevents this issue.
For reconstitution and daily use, keeping peptides on ice or at 4°C during working sessions maintains stability. Avoid leaving peptides at room temperature for extended periods during preparation.
Protecting from Light
Light exposure, particularly UV light, can cause photodegradation of peptide bonds and oxidation of sensitive amino acids. Always store peptides in their original dark amber vials or transfer to light-resistant containers.
If your peptides arrived in clear vials, transfer them to amber autosampler vials or wrap standard vials in aluminum foil. This simple step significantly extends shelf life for light-sensitive sequences.
During preparation and reconstitution, minimize exposure to direct light. Work in shaded areas or use amber containers during handling. This is especially important for peptides containing aromatic amino acids like phenylalanine, tyrosine, and tryptophan, which absorb UV light and are prone to photo-oxidation.
Avoiding Contamination
Contamination is one of the most common reasons peptides degrade before their time. Even trace amounts of moisture, bacteria, or foreign proteins can catalyze degradation pathways.
Always use sterile technique when handling peptides. This means:
- Working in a clean environment
- Using sterile vials and containers for storage
- Avoiding contact between the peptide powder or solution with non-sterile surfaces
- Using fresh, sterile syringes for each withdrawal if working with reconstituted solutions
For reconstituted peptides, bacterial growth can become significant within days to weeks, even when refrigerated. Most reconstituted peptide solutions are stable for 2-4 weeks at 4°C, but longer storage requires freezing. Some researchers add antimicrobial agents like benzyl alcohol to extend stability, though this requires careful consideration of compatibility with your intended use.
Never use peptide solutions that appear cloudy, have visible particles, or show signs of microbial growth. These are clear indicators of contamination that compromises both stability and safety.
